Patients testing positive for APC resistance or the Factor V Leiden mutation should be considered for testing for the FactorII (Prothrombin) G20210A variant. Individuals who are homozygous or heterozygous for the 20210A genotype have a 3-to 5-fold elevated risk for venous thrombosis.
DNA is isolated from whole blood and subjected to amplification (PCR). The test is performed on the Light Cycler instrument employing flurogenic reagents specific for the Factor V or Factor II mutations. Determination of the genotype is performed by melting curve analysis.
- Sample Required: 5 ml Whole Blood EDTA (purple top)
- Test Values: Homozygous normal (20210G); Heterozygous or Homozygous Abnormal (20210A)
- Analytic Time: Run weekly (Tuesdays) on samples received Wednesday through Monday noon; abnormal tests are confirmed by repeating tests.
