Ham’s Test

This test may be ordered in a patient with clinical features suggestive of paroxysmal nocturnal hemoglobinuria (PNH). In PNH, red cells that lack decay accelerating factor (DAF) will lyse in the presence of fresh normal serum. Flow cytometry studies have also been developed to aid in the diagnosis of PNH (currently not performed at Puget Sound Blood Center).

  • Sample Required: 7 mL fresh clotted sample or sodium citrate or ACD
  • Normal Test Values: Patients who do not have PNH will have a negative test.
  • Analytic Time: 24 hours if weekday, 72 hours if weekend. This test is not usually performed on a STAT basis.
  • Days Test is Set Up: Monday through Sunday
